Migration is one of the key processes of tumor cell proliferation, tumor cells need to have a certain ability to separate from the original tumor, and through the vascular wall, invade the surrounding normal tissue. Highly metastatic tumor cells often exhibit strong motility. A variety of substances can promote tumor cell migration, including tumor cell secretion factors, growth factors, extracellular matrix components (such as fibronectin and type IV collagen), and certain tumor metastasis target organ metabolites or secretions. These growth factors have a chemotactic effect on tumor cells and can make them undergo directional movement. The determination methods include cell scratch method and Transwell chamber method. The cell scratch assay is a convenient method to measure the migration and repair capacity of cells, similar to the in vitro wound healing model. In the culture dish or culture plate, use a micro gun or other hard objects to line the central area of cell growth, remove the central cells, and then continue to culture the cells to a set time, observe whether the surrounding cells grow to the central scratch area, so as to judge the migration ability of the cells. The experiment usually includes a normal control group and an experimental group, and the experimental group is a cell group after adding certain treatment factors, drugs or exogenous genes. By comparing the differences in the ability of different groups of cells to repair the scratch area, the migration and repair ability of each group of cells can be evaluated.
